Our FACSAria™ sample mixer, ARIA slide adaptor, calibration slide and user guides for the Becton Dickinson FACSAria, FACSVantage™, and Beckman Coulter MoFlo™ make it easy to add single cell molecular analysis to high throughput cell sorting.
FACS™ is a high throughput and extremely accurate front end for subsequent molecular analysis of single cells using our streamlined workflow. AmpliGrid adds single cell genophenotyping capabilities to the single cell immunophenotyping capabilities that are key to many immunology, stem cell, and oncology/hematology applications.
One key advantage of sorting single cells into the AmpliGrid slide rather than MTPs is its high placement accuracy of above 95%. In contrast, placement success into 96-well MTPs rarely exceeds 60-70% due to electrostatic deflection of cells before they reach the well bottoms. Moreover, since there is no straight-forward optical quality control (QC) option in tubes or MTPs, investigators are left to guess whether a failed PCR reaction was due to PCR failure, failed cell deposition, or whether there is a valid biological reason. AmpliGrid puts an end to this frustrating guess work. Due to its two-dimensional layout, correct cell placement of single cells can easily be verified visually before moving on to molecular analysis (also available as automated solution).
CellEvator is our sample mixer for the BD FACSAria™. Declining event counts or clogging often occur when sorting large or fragile cells, which tend to settle more quickly to the bottom of the sample tube, or adhere to the tube’s wall or to each other. Conventional mixing with a shaker or stir bar is often not effective in preventing declining event counts or clogging, because mixing is confined to the horizontal (x/y plane), while settlement is a vertical (z-axis) process. The CellEvator works by inducing surface waves (SAW) into the sample through the bottom of the sample tube; surface acoustic waves propagate vertically up the tube, thus off-setting settlement. Our patented SAW agitation allows for gentle, non-contact mixing that does not harm life cells. SAW mixing has been proven in the SlideBooster, Advalytix’ market leading line of active microarray hybridization stations, and the PlateBooster, our highly innovative contactless mixers for 96 and 384-well MTPS.
