Pre-amplifying single cells with the AmpliGrid system followed by standard qPCR combines existing workflows with superior single cell sensitivity.
The analysis of single cells using quantitative PCR often leads to unsatisfactory results due to the low gene copy number contained in a single cell sample. In addition, conventional single cell workflows are cumbersome, error-prone, and make inefficient use of precious template material.
In order to combine the easy-to-use workflows and single cell sensitivity of the AmpliGrid platform with quantitative PCR results, we developed a qPCR pre-amplification protocol. This protocol directly pre-amplifies single cells using the AmpliGrid 1µL PCR platform – no separate extraction step needed. The pre-amplified amplicons are then transferred onto a standard qPCR system for qPCR analysis.
Illustrating this qPCR protocol, our application report describes how to measure the mRNA expression of two genes in single human lymphocytes. This report was featured in the May email newsletter of www.gene-quantification.info/.
